Cloning and Characterization of a New Human CNA1 Transcript Variant

Calcineurin (CN) is the only Ca2+calmodulin (CaM) dependent protein serine/ threonine phosphatase consisting of a catalytic subunit, CNA, and a regulatory subunit, CNB. There are three subtypes of CNA, but CNAα is the most frequent one which is encoded by CNA1 gene. In the present study, a new variant of human CNA1 transcript, CNAα4, was isolated from a human fetal brain cDNA library. The CNAα4 lacks exon 2 of coding region and encodes a truncated protein of 454 amino acid residues. As a result, 67 amino acids are missed in the phosphatase catalytic domain. By in vitro binding assays, CNAα4 was found to have a comparable affinity to CaM as the largest isoform of CNAα, CNAα1. However, CNAα4 is more effective in activating nuclear factor of activated T cells, cytoplasmic 1 (NFATc1) than CNAα1, suggesting that the fragment of Ala20-Thr86 may be dispensable for the catalytic function of CNAα. On the contrary, this deletion in CNAα4 may favor the binding of CN to NFAT and then promote the activity of CN.
