The culture method for rat glomerular mesangial cells (MCs) in vitro was established.The effects of basic fibroblast growth factor (bFGF) on DNA synthesis of MC and expression of protooncogene c fos and c myc in MC were determined by 3H TdR incorporation,Dot blot and Northern blot analysis.The results showed that the value of 3H TdR incorporated into MC significantly increased when MCs were incubated with bFGF(20 ng/ml) for 18 hours ( P< 0 05),and reached peak at 24 hours;bFGF significantly induced the expression of protooncogene c fos c myc.The maximal expression of protooncogene c fos and c myc was respectively at 30 min and 1 hour after induced by bFGF.These data indicate that bFGF is a stronger mitogen for MC,the stimulation of bFGF for DNA synthesis of MC relates to the expression of protooncogene c fos and c myc induced by bFGF.
