摘要

用放射性配体[3H]SCH23390建立鸡视网膜多巴胺D1受体的放射性配体结合分析法,并测定不同试验组鸡视网膜多巴胺D1受体的Bmax值有显著性差异,KD值无明显差异.同时探讨了干,湿滤膜片对放射性测定结果的影响,以及影响试验结果的其它因素.实验表明湿滤膜片测量效果优于干滤膜片,膜受体即使贮存在-20℃条件下亦只能一周,否则影响受体的活性.

Abstract

The Dopamine D1 receptor in chicken retina in vitro was studied with [3H]SCH23390 (D1 selective antagonists). The radioligand binding assay of dopamine receptor in chicken retina was established; and the effect of dry and wet filters and other factors on the measurement of scintillation counters was studied. The results indicated that it is better to use wet filters for assay than to use'dry filter. The receptor in membrane should be kept at - 20℃ for only a week. The difference of Bmax was obvious in different groups but the KD was not obvious
