Errα Promotes Porcine Preadipocytes Adipogenesis by Inhibiting β-Catenin

Errα Promotes Porcine Preadipocytes Adipogenesis by Inhibiting β-Catenin

Abstract

Both estrogen-related receptor α (Errα) and Wnt/β-Catenin signaling pathways regulate adipogenesis. Studies have shown there is a crosstalk between Errα and Wnt/β-Catenin signaling pathways, and β-Catenin is the key factor of Wnt/β-Catenin signaling pathway. To study the relations between Errα and β-Catenin during adipogenesis, Ad-Errα virus was produced in 293A cells to infect porcine preadipocytes. LiCl and XCT790 were used for different treatments of porcine preadipocytes. Western blotting results showed that Errα protein level was escalated and β-Catenin protein level was decreased during adipogenesis. Microscopic observation showed that porcine preadipocytes were successfully infected by Ad-Errα by checking green fluorescence. Western blotting results showed that Ad-Errα stimulated Errα protein expression, and XCT790 suppressed Errα protein expression in the porcine preadipocytes. Oil-red-O staining showed that β-Catenin suppressed adipogenesis while Errα promoted adipogenesis by suppressing β-Catenin. Furthermore, Western blotting results suggested that LiCl stabilized β-Catenin expression, and Errα suppressed β-Catenin expression during porcine preadipocyte adipogenesis. These findings imply that Errα promotes adipogenesis by inhibiting β-Catenin expression.
