Does Death-associated Protein Kinase(DAPK) Negatively or Positively Regulate Autophagy?

Abstract

Autophagy is a regulated catabolic pathway by which mammalian cells degrade and recycle macromolecules and organelles. Recently, autophagy was demonstrated as an adaptive mechanism to protect organisms against diverse pathological conditions, including infection, cancer, neurodegeneration, aging, and heart disease. By removing protein aggregates, as well as damaged or excess organelles, autophagy plays critical roles in maintaining cellular energy homeostasis and the cell adaptation to environmental stresses. A prodeath effect was found when autophagy is insufficient or excessive. The dynamic of autophagic activation is precisely regulated by the network of autophagy signaling pathways. Deathassociated protein kinase(DAPK) is a multidomain stressregulated protein kinase in a wide range of signaling pathways, including apoptosis, autophagy and membrane vacuolation. In this review, we summarized the evidence of DAPK to negatively or positively regulate autophagy under different conditions.
