Abstract:Aeromonas hydrophila, a common pathogen, can cause disease in freshwater fish known as “Hemorrhagic Septicemia”, “Ulcer Disease” or “Red-Sore Disease”, etc. To reveal the antibacterial effects of traditional Chinese medicines (TCMs) on A. hydrophila, the ethanol-extracts of Rhei Radix et Rhizoma (RRR, root or stem of Rheum officinale Baill), Glycyrrhizae Radix et Rhizoma (GRR, root or stem of Glycyrrhiza uralensis Fisch) and Lonicerae Japonicae Flos (LJF, flower of Lonicera Japonica Thunb.) were evaluated. The results showed the minimum inhibitory concentrations (MIC) values of RRR, GRR and LJF for the A. hydrophila were 1.0 mg/mL, 7.8 mg/mL and 3.9 mg/mL, respectively. The minimal bactericidal concentration (MBC) values of them were 4.0 mg/mL, 31.2 mg/mL and 7.8 mg/mL, respectively. After analysis by SDS-PAGE, two-dimensional electrophoresis (2-DE) and matrix+assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF-TOF-MS), the protein levels of 30S ribosomal protein S1 (S1), F0F1 ATP synthase β subunit (ATPase-β), isocitrate lyase and enolase (NSE) of A. hydrophila could be induced by TCMs. Furthermore, the RT-qPCR results showed that the mRNA levels of these four genes were affected by TCMs. These results were in good agreement with TCMs applications in aqueous infections, and suggested that the inhibition of ethanol-extracts in A. hydrophila could be achieved through regulating the expression levels of metabolic proteins.
