Abstract:Nuclear RNA are extruded from the cytoplasm of rat apoptotic thymocytes. RNA was extracted from the culture supernatant of murine splenocytes following apoptosis induction. The RNA content of apoptotic cells was less than that of living cells as determined in FACS analysis. Methyl green pyronin Y staining showed that RNA existed in extracellular space of mouse spleen, indicating that RNA was also extruded from spleen cells in vivo . The ratio of supernatant RNA to total spleen RNA was used to estimate the quantity of RNA extruded by apoptotic cells. The ratio of supernalant RNA to total RNA in BALB/c mice decreased with aging (72% for 3 weeks, 58.4% for 3 months and 45% for 6 months), while that of the BXSB mice of different ages remained at 75%-83%. RNase activity in the spleen supernatant, as measured by single radial enzyme diffusion assays, of 6 month old BXSB mice was significantly lower than that of BALB/c mice ( P <0.05), although no significant difference was not observed between BALB/c and BXSB mice of younger age.
