Abstract:To avoid the degradation of nucleic acids is critical for the preservation of collected clinical liver tissue samples for subsequent experimental analyses. This study evaluated the RNA integrity in excised mouse liver tissues when placed at room temperature for different periods, thus to provide data for the collection and preservation protocols of clinical liver samples. The extracted total RNA was kept at room temperature for 0 to 180 min, and the integrity of RNA was examined by using agarose gel electrophoresis, RT-PCR and bioanalyzer assays. No significant RNA degradation was observed by electrophoresis and the RT-PCR analyses to probe for β-actin, GAPDH and Trp53. The bioanalyzer analyses showed that the RNA integrity number (RIN) of all samples were greater than 7.9 as qualified for further analyses. We concluded that up to 3 hours at room temperature storage of the excised liver tissues resulted in no significant RNA integrity loss.
